Adult Dosing
- 200 mg orally 2 to 3 times daily
- Maximum daily dose: 600 mg
- Administer with food or milk to minimize gastrointestinal irritation
Pediatric Dosing
- Safety and efficacy in pediatric patients not established
Indications
- Treatment of rheumatoid arthritis
- Treatment of osteoarthritis
- Management of acute musculoskeletal pain and inflammation
Mechanism of Action
- Non-steroidal anti-inflammatory drug (NSAID)
- Inhibits cyclooxygenase (COX-1 and COX-2) enzymes, reducing prostaglandin synthesis
Contraindications
- Known hypersensitivity to clofezone, phenylbutazone, or other NSAIDs
- Active peptic ulcer disease or gastrointestinal bleeding
- Severe hepatic or renal impairment
- Severe heart failure
- History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
Adverse Reactions
- Gastrointestinal distress, dyspepsia, nausea, abdominal pain
- Peptic ulceration and gastrointestinal bleeding
- Dizziness, headache, drowsiness
- Fluid retention, edema, hypertension
- Elevated liver enzymes, rare hepatotoxicity
- Hypersensitivity reactions, rash, pruritus
Drug Interactions
- Anticoagulants (e.g., warfarin) and antiplatelet agents: increased risk of bleeding
- Other NSAIDs or corticosteroids: increased risk of gastrointestinal ulceration and adverse effects
- Antihypertensives, ACE inhibitors, and diuretics: decreased antihypertensive efficacy and increased nephrotoxicity risk
- Lithium and methotrexate: reduced renal clearance leading to potential toxicity
