Forms & Strengths
- Tablet: 50 mg
- Tablet: 150 mg
Adult Dosing
- 50 mg to 150 mg orally 3 times daily
- Dose adjusted based on individual patient therapeutic response and tolerability
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
- Not recommended for use in children
Indications
- Symptomatic treatment of spasticity in adults following neurological diseases
- Treatment of painful muscle spasms associated with musculoskeletal conditions
Mechanism of Action
- Centrally acting skeletal muscle relaxant
- Exerts membrane-stabilizing effect on primary afferent fibers and motor neurons
- Inhibits spinal reflex arcs and exhibits dose-dependent inhibition of voltage-gated sodium and calcium channels
Contraindications
- Hypersensitivity to tolperisone or its excipients
- Myasthenia gravis
- Breastfeeding
Adverse Reactions
- Muscular weakness
- Hypotension
- Nausea and dyspepsia
- Headache and dizziness
- Rarely: hypersensitivity reactions including anaphylaxis
Drug Interactions
- Enhances effects of other centrally acting muscle relaxants, requiring dose reduction
- Potentiates effects of niflumic acid and other NSAIDs; consider dose reduction when co-administered
- CYP2D6 and CYP2C19 substrates; potential for pharmacokinetic interactions with inhibitors or inducers
