Forms & Strengths
- Injectable solution: 25 mg/mL
Adult Dosing
- Subcutaneous or intramuscular: 25-50 mg every 4-6 hours as needed for pain
- Intravenous: 5-10 mg slowly, titrated to clinical response
Pediatric Dosing
- Safety and efficacy in pediatric patients under 12 years have not been established; use is generally not recommended
Indications
- Management of moderate to severe pain
- Analgesic supplement during anesthesia
Mechanism of Action
- Synthetic phenylpiperidine opioid agonist acting primarily at mu-opioid receptors in the central nervous system, inhibiting ascending pain pathways and altering perception of and response to pain
Contraindications
- Hypersensitivity to anileridine or other opioids
- Significant respiratory depression
- Acute or severe bronchial asthma in an unmonitored setting
- Known or suspected paralytic ileus
Adverse Reactions
- Respiratory depression
- Sedation and somnolence
- Hypotension and orthostatic hypotension
- Nausea and vomiting
- Constipation
- Dizziness and lightheadedness
Drug Interactions
- Central nervous system depressants (alcohol, barbiturates, benzodiazepines, general anesthetics) increase risk of profound sedation, respiratory depression, coma, and death
- Monoamine oxidase inhibitors (MAOIs) may precipitate severe unpredictable hyperactive or hypotensive crises
- Mixed agonist/antagonist opioid analgesics may reduce analgesic effect or precipitate withdrawal symptoms
