Forms & Strengths
- Intravenous solution: 50 mg/mL
Adult Dosing
- General anesthesia induction: 5-10 mg/kg IV administered over 30-60 seconds
- Short surgical procedures: 5-7 mg/kg IV
Pediatric Dosing
- Safety and efficacy in pediatric patients not well established; use generally avoided or restricted based on clinical trial data
Indications
- Induction of general anesthesia
- Short-duration surgical procedures
- Supplementation of regional anesthesia
Mechanism of Action
- Ultra-short-acting non-barbiturate general anesthetic agent
- Depresses the central nervous system through potentiation of GABAergic neurotransmission and reduction of reticular activating system activity
Contraindications
- Hypersensitivity to propanidid or its excipients
- Severe cardiovascular disease or shock
- Epilepsy or history of seizures
- Porphyria
Adverse Reactions
- Transient hyperventilation followed by respiratory depression or apnea
- Hypotension and tachycardia due to histamine release and myocardial depression
- Involuntary muscle movements, tremors, and twitching
- Anaphylactoid and severe hypersensitivity reactions
- Thrombophlebitis at the injection site
Drug Interactions
- Potentiation of central nervous system depression when co-administered with opioids, sedatives, or other general anesthetics
- Enhanced hypotensive effect when combined with antihypertensive agents or vasodilators
- Prolonged neuromuscular blockade when used concomitantly with non-depolarizing muscle relaxants
