bezitramide
N02AC05
Adult Dosing
- Initial: 5 to 10 mg orally every 4 to 6 hours as needed for severe pain
- Titrate dose based on analgesic response and tolerance
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
- Use generally not recommended in children
Indications
- Management of moderate to severe acute and chronic pain
Mechanism of Action
- Full agonist at mu-opioid receptors in the central nervous system
- Inhibits ascending pain pathways, altering the perception of and response to pain
Contraindications
- Hypersensitivity to bezitramide
- Significant respiratory depression
- Acute or severe bronchial asthma in an unmonitored setting
- Known or suspected gastrointestinal obstruction, including paralytic ileus
Adverse Reactions
- Respiratory depression
- Sedation and somnolence
- Constipation
- Nausea and vomiting
- Dizziness and headache
- Hypotension and orthostatic hypotension
- Urinary retention
- Physical dependence and tolerance
Drug Interactions
- Central nervous system (CNS) depressants (alcohol, benzodiazepines, other opioids, sedatives) increase risk of profound sedation, respiratory depression, coma, and death
- Mixed agonist/antagonist or partial agonist opioid analgesics may reduce analgesic effect or precipitate withdrawal symptoms
- Monoamine oxidase inhibitors (MAOIs) may cause severe adverse reactions including unpredictable potentiation of opioid effects
