Forms & Strengths
- Oral tablets: 1 mg
- Oral tablets: 2 mg
Adult Dosing
- Initial dose: 1 mg to 2 mg daily in divided doses
- Maintenance dose: Titrated based on clinical response, typically up to 4 mg to 6 mg daily
- Maximum daily dose: 6 mg daily
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Treatment of Parkinson's disease
- Management of drug-induced extrapyramidal symptoms
Mechanism of Action
- Central anticholinergic agent
- Blocks central muscarinic receptors to help restore the balance between cholinergic and dopaminergic activity in the basal ganglia
Contraindications
- Hypersensitivity to mazaticol
- Angle-closure glaucoma
- Gastrointestinal obstruction, paralytic ileus, or toxic megacolon
- Myasthenia gravis
- Severe urinary tract obstruction due to prostatic hyperplasia
Adverse Reactions
- Dry mouth
- Blurred vision
- Constipation
- Urinary retention
- Drowsiness or dizziness
- Tachycardia
- Cognitive impairment or confusion in elderly patients
Drug Interactions
- Enhanced anticholinergic adverse effects when co-administered with other anticholinergic agents (e.g., tricyclic antidepressants, antihistamines)
- May antagonize the gastrointestinal effects of prokinetic agents (e.g., metoclopramide)
