Adult Dosing
- Depression: 100 mg to 300 mg daily in divided doses
- Max recommended dose: 300 mg daily
Pediatric Dosing
- Safety and efficacy not established in pediatric patients
- Not recommended for use in children
Indications
- Major depressive disorder
- Apathetic depression in elderly patients
Mechanism of Action
- Atypical antidepressant with weak monoamine oxidase inhibitory properties
- Enhances dopaminergic and serotonergic neurotransmission
- Exhibits anticholinergic and antihistaminergic effects
Contraindications
- Hypersensitivity to minaprine
- Acute myocardial infarction
- Severe hepatic impairment
- History of epilepsy or convulsive disorders
- Concomitant use with MAO inhibitors
Adverse Reactions
- Insomnia
- Anxiety and agitation
- Dizziness and headache
- Dry mouth and gastrointestinal disturbances
- Seizures in overdose or predisposed patients
- Liver enzyme elevations
Drug Interactions
- Monoamine Oxidase Inhibitors (MAOIs): risk of hypertensive crisis and central toxicity
- Central nervous system depressants: enhanced sedative effects
- Anticholinergic agents: additive anticholinergic adverse effects
- Levodopa: potential pharmacodynamic antagonism or potentiation
