Forms & Strengths
- Tablets: 12.5mg, 20mg, 30mg, 40mg
- Extended-release tablets: 20mg, 40mg
- Oral solution: 10mg/5mL
Adult Dosing
- Initial dose: 12.5mg twice daily, titrate to 20-40mg twice daily
- Maximum dose: 80mg/day in divided doses
- Elderly: initiate with 12.5mg once daily, titrate to 20-30mg/day in divided doses
Pediatric Dosing
- Not approved for use in pediatric patients under 18 years old
Indications
- Major depressive disorder
- Anxiety disorders
Mechanism of Action
- Selective serotonin 5-HT1A receptor partial agonist
- Antagonist activity at alpha-1 adrenergic receptors
Contraindications
- Known hypersensitivity to gepirone
- Concomitant use of monoamine oxidase inhibitors (MAOIs)
- Concomitant use of potent CYP3A4 inhibitors
Adverse Reactions
- Dizziness
- Nausea
- Headache
- Somnolence
- Dry mouth
- Constipation
Drug Interactions
- Increased risk of serotonin syndrome with concomitant use of serotonergic agents
- Concomitant use with CYP3A4 inhibitors may increase gepirone levels
- Gepirone may increase levels of concomitantly administered CYP2D6 substrates
