Forms & Strengths
- Injection: 100 mg antimony/mL in 100 mL vials
Adult Dosing
- Visceral leishmaniasis: 20 mg/kg/day IV or IM for 28-30 days
- Cutaneous leishmaniasis: 20 mg/kg/day IV or IM for 20 days
Pediatric Dosing
- Visceral leishmaniasis (children >= 1 month): 20 mg/kg/day IV or IM for 28-30 days
- Cutaneous leishmaniasis (children >= 1 month): 20 mg/kg/day IV or IM for 20 days
Indications
- Treatment of leishmaniasis (visceral, cutaneous, and mucosal)
Mechanism of Action
- Pentavalent antimonial prodrug converted to trivalent antimony, inhibiting parasite glycolysis and fatty acid oxidation
Contraindications
- Hypersensitivity to antimonials
- Severe renal impairment
- Severe hepatic impairment
- Significant cardiac disease or QTc prolongation
Adverse Reactions
- Electrocardiographic changes (QT prolongation, T-wave inversion)
- Myalgia and arthralgia
- Pancreatitis
- Elevated transaminases
- Nausea, vomiting, and abdominal pain
- Injection site pain and thrombophlebitis
Drug Interactions
- QT-prolonging drugs (increased risk of fatal arrhythmias)
- Cardiotoxic medications (additive toxicity)
- Nephrotoxic drugs (increased risk of renal dysfunction)
