Adult Dosing
- Onchocerciasis: 8 mg (four 2-mg tablets) as a single, one-time oral dose
Pediatric Dosing
- Pediatric patients aged 12 years and older: 8 mg as a single, one-time oral dose
- Safety and efficacy in children under 12 years of age have not been established
Indications
- Treatment of onchocerciasis (river blindness) caused by Onchocerca volvulus in patients aged 12 years and older
Mechanism of Action
- Macrocyclic lactone that binds to glutamate-gated chloride channels in invertebrate nerve and muscle cells
- Increases membrane permeability to chloride ions, resulting in hyperpolarization, paralysis, and death of the parasite (microfilariae)
Contraindications
- Hypersensitivity to moxidectin or any of the formulation excipients
Adverse Reactions
- Eosinophilia
- Pruritus
- Rash
- Headache
- Dizziness
- Tachycardia
- Hypotension
- Myalgia
- Arthralgia
- Mazzotti-type reactions due to microfilarial lysis (e.g., edema, lymphadenopathy)
Drug Interactions
- No formal clinical drug-interaction studies have been conducted
- Potential for additive neurotoxicity or pharmacokinetic alterations with strong inhibitors or inducers of CYP3A4, though moxidectin is primarily eliminated unchanged or via minor pathways
