Adult Dosing
- Nausea and vomiting, motion sickness: 25 to 50 mg orally daily in divided doses
- Appetite stimulation (off-label): 25 to 50 mg orally 1 to 3 times daily before meals
Pediatric Dosing
- Safety and efficacy not well established in children under 12 years
- Motion sickness (children >= 12 years): 25 mg orally daily or divided doses
Indications
- Prevention and treatment of nausea, vomiting, and dizziness associated with motion sickness
- Appetite stimulant in various cachectic states (off-label)
Mechanism of Action
- Antihistamine with central anticholinergic and local anesthetic properties
- Depresses labyrinthine excitability and conduction in the middle ear vestibular pathway
- Blocks H1 receptors in the chemoreceptor trigger zone to suppress nausea and vomiting
Contraindications
- Hypersensitivity to buclizine or other piperazine derivatives (e.g., cetirizine, meclizine)
- Angle-closure glaucoma
- Symptomatic prostatic hypertrophy
- Lower respiratory tract symptoms including asthma
Adverse Reactions
- Drowsiness and sedation
- Dry mouth, nose, and throat
- Blurred vision
- Urinary retention
- Constipation
- Paradoxical excitation in children
Drug Interactions
- Central nervous system (CNS) depressants (e.g., alcohol, barbiturates, opioids, sedatives) enhance sedative effects
- Anticholinergic drugs (e.g., tricyclic antidepressants, MAO inhibitors) may potentiate anticholinergic adverse effects
