Forms & Strengths
- Tablet: 10 mg
- Chewable Tablet: 5 mg
- Syrup: 1 mg/mL
- Rapid-disintegrating tablet (ODT): 10 mg
Adult Dosing
- Allergic rhinitis and urticaria: 10 mg orally once daily
- Hepatic impairment (Child-Pugh Class A, B, or C) or severe renal impairment (GFR < 30 mL/min): 10 mg orally every other day as initial dose
Pediatric Dosing
- Children 2 to 5 years: 5 mg orally once daily (syrup or chewable)
- Children 6 years and older: 10 mg orally once daily
Indications
- Relief of symptoms associated with seasonal allergic rhinitis
- Treatment of chronic idiopathic urticaria
Mechanism of Action
- Long-acting, selective peripheral histamine H1-receptor antagonist
- Lacks significant central nervous system sedative and anticholinergic properties at recommended doses
Contraindications
- Hypersensitivity to loratadine or any component of the product
Adverse Reactions
- Headache
- Somnolence
- Fatigue
- Dry mouth
- Nausea
Drug Interactions
- CYP3A4 and CYP2D6 inhibitors (e.g., ketoconazole, erythromycin, cimetidine) may increase plasma concentrations of loratadine and its active metabolite desloratadine, though usually without clinical significance
- Increased sedation or CNS depression when co-administered with other CNS depressants or alcohol (despite its non-sedating profile)
