Forms & Strengths
- Intravitreal injection: 2.25 mg/0.25 mL in single-use vial
Adult Dosing
- Cytomegalovirus (CMV) retinitis: Induction phase: 3.3 mg (0.37 mL) injected intravitreally once every other week for 2 doses
- Cytomegalovirus (CMV) retinitis: Maintenance phase: 3.3 mg (0.37 mL) injected intravitreally once every 4 weeks
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Local treatment of cytomegalovirus (CMV) retinitis in patients with acquired immunodeficiency syndrome (AIDS) who are intolerant of or unresponsive to other treatments
Mechanism of Action
- Antisense oligonucleotide that inhibits human cytomegalovirus (CMV) replication by binding to complementary sequences on viral mRNA, preventing protein synthesis
Contraindications
- Hypersensitivity to fomivirsen or any component of the formulation
- Recent (within 2-4 weeks) treatment with systemic or ocular cidofovir due to risk of severe ocular inflammation
Adverse Reactions
- Ocular: Intraocular pressure increased, vitritis, uveitis, vision abnormal, cataract, retinal detachment, retinal hemorrhage, eye pain
- Systemic: Fever, chills, abnormal liver function tests, asthenia, headache, nausea, pneumonia
Drug Interactions
- Cidofovir: Concomitant administration with systemic or intravitreal cidofovir is contraindicated due to an increased risk of severe ocular inflammation (iritis/uveitis)
- Other intravitreal agents: Potential additive ocular toxicity when used concurrently with other intraocular drugs
