pranoprofen
S01BC09
Adult Dosing
- Instill 1 to 2 drops into the affected eye 4 times daily
- Duration of therapy depends on clinical response, typically up to 2 weeks for inflammatory conditions
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been firmly established
- Use in children requires careful risk-benefit assessment by a specialist
Indications
- Treatment of inflammatory conditions of the external eye and anterior segment
- Examples include blepharitis, conjunctivitis, keratitis, and scleritis
- Post-operative inflammation following ocular surgery
Mechanism of Action
- Non-steroidal anti-inflammatory drug (NSAID) with pronounced anti-inflammatory, analgesic, and antipyretic properties
- Inhibits cyclooxygenase (COX-1 and COX-2), thereby blocking the biosynthesis of prostaglandins responsible for ocular inflammation, pain, and miosis
Contraindications
- Hypersensitivity to pranoprofen or any component of the formulation
- History of hypersensitivity, asthma, or allergic reactions to aspirin or other NSAIDs
- Active acute viral, bacterial, or fungal infections of the eye lacking anti-infective treatment
Adverse Reactions
- Transient stinging, burning, or irritation upon instillation
- Blurred vision or visual discomfort
- Ocular hyperemia, pruritus, or allergic conjunctivitis
- Rarely, corneal epithelial breakdown or keratitis with prolonged use
Drug Interactions
- Concurrent use of topical ophthalmic corticosteroids may delay corneal healing and increase the risk of infection
- Additive adverse effects possible with other topical NSAIDs
- Systemic absorption is minimal, making significant pharmacokinetic drug interactions unlikely
