latanoprost and dorzolamide
S01EE52
Forms & Strengths
- Ophthalmic solution: latanoprost 0.005% + dorzolamide 2% (combination)
Adult Dosing
- One drop in the affected eye(s) once daily in the evening
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Reduction of elevated intraocular pressure in patients with open-angle glaucoma or ocular hypertension inadequately controlled by monotherapy
Mechanism of Action
- Latanoprost is a selective FP prostanoid receptor agonist that increases uveoscleral outflow of aqueous humor
- Dorzolamide is a topical carbonic anhydrase II inhibitor that decreases aqueous humor secretion by the ciliary body epithelium
Contraindications
- Hypersensitivity to latanoprost, dorzolamide, or any component of the formulation
- History of sulfonamide allergy (due to systemic absorption of dorzolamide)
Adverse Reactions
- Eye irritation, burning, stinging, and conjunctival hyperemia
- Blurred vision and superficial punctate keratitis
- Increased iris pigmentation and eyelash changes
- Bitter taste (dysgeusia) associated with dorzolamide
Drug Interactions
- Concomitant use of two topical prostaglandins or analogs is not recommended due to potential paradoxical elevation of IOP
- Additive systemic effects when used concurrently with oral carbonic anhydrase inhibitors
- Precipitation may occur if eyedrops containing thimerosal are instilled simultaneously; separate administration by at least 5 minutes
