mangafodipir
V08CA05
Forms & Strengths
- Intravenous solution: 0.01 mmol/mL (manganese carbohydrate complex)
Adult Dosing
- Diagnostic imaging: 5 µmol/kg (0.5 mL/kg) administered as a slow intravenous injection or infusion
- Administered approximately 10 to 24 minutes prior to magnetic resonance imaging (MRI) of the liver
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established
Indications
- Contrast agent for magnetic resonance imaging (MRI) to assist in the detection of focal liver lesions
Mechanism of Action
- Paramagnetic contrast agent containing manganese, which shortens T1 relaxation times of protons in hepatic tissue
- Selectively taken up by normal hepatocytes, enhancing the contrast between normal liver parenchyma and non-functioning lesions (e.g., metastases, cysts)
Contraindications
- Known hypersensitivity to mangafodipir or manganese-containing compounds
- Severe renal impairment (though primarily hepatobiliary eliminated, caution is advised)
- Severe hepatic failure where hepatocyte uptake is absent
Adverse Reactions
- Transient flushing
- Injection site pain or warmth
- Nausea
- Headache
- Mild, transient decreases in blood pressure
Drug Interactions
- No formal well-controlled pharmacokinetic drug interaction studies have been conducted
- Potential for additive hemodynamic effects with antihypertensive agents due to transient vasodilation
