Forms & Strengths

Adult Dosing
- Single intravenous injection of 111 to 222 MBq (3 to 6 mCi) for single-photon emission computed tomography (SPECT) imaging of dopamine D2 receptors in the brain
Pediatric Dosing
- Safety and efficacy in pediatric patients have not been established; use is generally restricted to adults
Indications
- Diagnostic visualization of striatal dopamine D2 receptors using SPECT imaging
- Evaluation of neurodegenerative movement disorders such as Parkinson's disease, multiple system atrophy, and progressive supranuclear palsy
Mechanism of Action
- Iodine-123 iolopride is a radiolabeled benzamide derivative that binds with high affinity and selectivity to striatal dopamine D2 receptors
- Gamma emissions from the iodine-123 radioisotope are detected externally via SPECT to quantify receptor density and functional integrity
Contraindications
- Known hypersensitivity to iodine-123 or any component of the formulation
- Pregnancy and lactation due to fetal and infant radiation risks
Adverse Reactions
- Rare transient headache, dizziness, or flushing associated with injection
- Potential hypersensitivity or allergic reactions ranging from mild rash to anaphylaxis
- Radiation exposure associated with diagnostic radiopharmaceuticals
Drug Interactions
- Concomitant administration of strong dopamine D2 receptor antagonists (e.g., antipsychotics, metoclopramide) competitively inhibits binding and alters imaging results
- Dopamine-depleting agents (e.g., tetrabenazine) may affect receptor occupancy and uptake measurements
