flutemetamol (18F)
V09AX04
Forms & Strengths
- Single‑use sterile vial
- Fluorine‑18 labeled flutemetamol
- Activity 185–370 MBq (5–10 mCi) per dose
- Prepared in saline for IV injection
Adult Dosing
- IV bolus injection of 185–370 MBq (5–10 mCi) over ≤5 seconds
- Inject 10 minutes before PET acquisition; imaging starts 90 minutes post‑injection
- Adjust activity for patient weight >90 kg (up to 555 MBq)
Pediatric Dosing
- Not approved for use in patients <18 years
- Safety and dosing not established in pediatric population
Indications
- Detection of cerebral amyloid plaques to aid diagnosis of Alzheimer’s disease
- Differentiation of Alzheimer’s disease from other dementias
- Research use in clinical trials of anti‑amyloid therapies
Mechanism of Action
- Fluorine‑18 labeled analogue of thioflavin that binds with high affinity to β‑amyloid aggregates
- After IV administration, crosses blood‑brain barrier and localizes to amyloid plaques
- PET detects emitted positrons, providing a map of plaque distribution
Contraindications
- Known hypersensitivity to flutemetamol or any excipients
- Pregnancy (radiation exposure to fetus)
- Breast‑feeding (radiation to infant)
Adverse Reactions
- Mild headache
- Nausea or vomiting
- Transient injection‑site pain or erythema
- Dizziness or feeling of warmth
- Rare allergic reaction (urticaria, bronchospasm)
Drug Interactions
- No clinically significant drug‑drug interactions reported
- Concurrent administration of other PET radiopharmaceuticals may require spacing to avoid cross‑contamination
- Avoid high‑dose iodinated contrast within 24 h if brain PET is planned (minimal effect)
