Forms & Strengths
- Injectable suspension: 1.85 GBq/mL (50 mCi/mL) at calibration time
- Radioactive diagnostic/therapeutic agent supplied in single-dose glass vials
Adult Dosing
- Palliative treatment of bone pain: 37 MBq/kg (1.0 mCi/kg) body weight administered via slow intravenous injection over 1 minute
Pediatric Dosing
- Safety and efficacy in pediatric patients below the age of 18 years have not been established
Indications
- Palliative treatment of bone pain in patients with osteoblastic bone metastases
Mechanism of Action
- Beta-emitting radioisotope that localizes in areas of active osteoblastic bone formation, delivering targeted ionizing radiation to relieve metastatic bone pain
Contraindications
- Hypersensitivity to samarium or phosphonate compounds
- Pregnancy and lactation
- Severe bone marrow suppression unless deemed medically necessary
Adverse Reactions
- Transient myelosuppression (leukopenia and thrombocytopenia)
- Nausea and vomiting
- Pain flare shortly after administration
- Injection site reactions
Drug Interactions
- Calcitonin or other bone metabolism modifiers may theoretically alter uptake of samarium 153Sm hydroxyapatite
- Concurrent chemotherapy or external beam radiation therapy increases the risk of severe bone marrow suppression
